چکیده

We investigated the effects of 1 and 3 mM 2,3-butanedione monoxime (BDM, diacetyl monoxime) on excitation and contraction of cardiac muscle in several types of preparations at various levels of organization. We selected a concentration of BDM that was not expected to affect sarcolemmal calcium flux and action potential duration in cardiac tissue. Two indicators were used to record intracellular calcium. Aequorin, a bioluminescent calcium indicator, was used in studies with ferret papillary muscle preparations, and fura-2, a fluorescent calcium indicator, was used in studies with guinea pig cardiac myocytes. In both cases, addition ofBDM resulted in a reduction of peak intracellular calcium released from the sarcoplasmic reticulum and a reduction of peak twitch force. The duration of the action potential of isolated myocytes

Preservation of myocyte contractile function after hypothermic, hyperkalemic cardioplegic arrest with 2, 3-butanedione monoxime.

One proposed contributory mechanism for depressed ventricular performance after hypothermic, hyperkalemic cardioplegic arrest is a reduction in myocyte contractile function caused by alterations in intracellular calcium homeostasis. Because 2,3-butanedione monoxime decreases intracellular calcium transients, this study tested the hypothesis that 2,3-butanedione monoxime supplementation of the h...

2,3-Butanedione monoxime increases speed of relaxation in single muscle ®bres of frog

The effects of 2,3-butanedione monoxime (BDM) on intracellular Ca transient and cross-bridge function were studied in frog single ®bres from the anterior tibialis muscle of Rana temporaria (sarcomere length, 2.2 lm; temperature, 2±4 °C). The ̄uorescent dye ̄uo-3 was used to monitor the intracellular free calcium concentration ([Ca]i) during isometric contractions. BDM (1±5 mM) reduced the ampli...

2,3-Butanedione monoxime increases speed of relaxation in single muscle fibres of frog.

The effects of 2,3-butanedione monoxime (BDM) on intracellular Ca2+ transient and cross-bridge function were studied in frog single fibres from the anterior tibialis muscle of Rana temporaria (sarcomere length, 2.2 microm; temperature, 2-4 degrees C). The fluorescent dye fluo-3 was used to monitor the intracellular free calcium concentration ([Ca2+]i) during isometric contractions. BDM (1-5 mM)...

Deactivation of contraction by quick releases in the isolated papillary muscle of the cat. Effects of lever damping, caffeine, and tetanization.

In the isolated cat papillary muscle, the time course of activation during isometric contraction was measured using instantaneous velocity of shortening after quick releases at constant contractile element length and controlled load. Activation rose and declined ahead of isometric force. This course of activation could be altered by the mode of the quick release. Undamped quick releases increas...
